Abstract:
OBJECTIVE To observe the impacts of electroacupuncture on the circadian rhythm of blood pressure, the protein and transcription expression of RORα and BMAL1 of the heart and hypothalamus of spontaneously hypertensive rats, and explore the mechanism of electroacupuncture in treating disturbance of circadian rhythm of blood pressure.
METHODS Twenty 16-week-old spontaneously hypertensive rats were selected and acclimatized for one week before being randomly assigned to four groups: model group, electroacupuncture group, sham electroacupuncture group, and western medicine group. Additionally, 5 age-matched Wistar-Kyoto (WKY) rats were selected as a control group. Acupuncture intervention was administered at the bilateral "Neiguan" (PC6) and "Zusanli" (ST36) acupoints, as well as the "Baihui" (GV20) acupoint. The 24-hour ambulatory blood pressure and body weight of rats were measured before and after the intervention; blood pressure was recorded every 4 hours, with ZT0 defined as 08:00 (the time the animal facility lights were turned on). Immunohistochemistry, immunofluorescence, and Western blot were used to observe the protein expression of RORα and BMAL1 in the heart and hypothalamus, while quantitative real-time PCR (qRT-PCR) was used to detect the transcriptional levels of RORα and BMAL1 in these tissues.
RESULTS Compared with the model group, the blood pressure in the electroacupuncture group was low in light and high at dark, the dipper ratio was increased (P<0.001), and the expression of RORα and BMAL1 in the heart and hypothalamus were all increased (P<0.05,P<0.001,). In the sham electroacupuncture group, the blood pressure could not reach the normal level of circadian oscillation and there were no significant differences in the expression of RORα and BMAL1 in the heart and hypothalamus. In the western medicine group, the level of nocturnal rhythmic oscillations of blood pressure was better than that in the model group, but no significant difference was seen in the dipper rate, the expression of RORα protein in the heart and hypothalamus were both elevated (P<0.001), and the hypothalamus BMAL1 protein and transcription levels were significantly elevated (P<0.01,P<0.001).
CONCLUSION Electroacupuncture can improve the circadian rhythm disorder of blood pressure in hypertensive rats, which may be related to regulate the circadian rhythm system by up-regulating the expression of RORα and BMAL1 in the heart and hypothalamus.
